目的研究大孔树脂纯化山香圆叶中总黄酮的工艺条件。方法采用静态吸附法筛选6种大孔树脂,再用动态吸附法优化山香圆叶中总黄酮的纯化工艺。结果山香圆叶中总黄酮的最佳纯化工艺为:上样浓度为0.1 g·m L-1(以生药计),上样速率为2 BV·h-1,上样体积为2 BV,用2 BV去离子水洗脱、5 BV 50%乙醇以2 BV·h-1的速率洗脱;在此工艺下,总黄酮的转移率为60.48%,产物中总黄酮的纯度为41.11%。结论 HPD400型大孔树脂对山香圆叶中总黄酮有良好的纯化效果。
Objective To study the conditions for the purification of total flavonoids from Shannon-Ylang Yuan with macroporous resin. Methods Six kinds of macroporous resins were screened by static adsorption method and the purification process of total flavonoids in Shanxiang circular leaves was optimized by dynamic adsorption method. Results The optimal purification technology of total flavonoids in Shannon leaves was as follows: loading concentration was 0.1 g · m L-1 (crude drug), loading rate was 2 BV · h-1, loading volume was 2 BV, 2 BV eluting with 5 BV 50% ethanol at a rate of 2 BV · h-1. Under this process, the total flavonoid transfer rate was 60.48% and the total flavone purity in the product was 41.11%. Conclusion HPD400 macroporous resin has good purification effect on the total flavonoids in Shanxiang round leaves.
