目的观察“通利大肠”对慢性阻塞性肺疾病(COPD)模型大鼠外周血及肺泡灌洗液中白介素-8(IL-8)、白介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)和白介素-10(IL-10)含量变化,从炎症调控角度探讨COPD“从肠论治”的效应机制。方法用气管注射脂多糖加熏香烟联合造模方法建立COPD大鼠模型,随机分为正常组、模型组、治肠组、治肺组及肺肠同治组。正常组、模型组灌胃纯净水,各给药组灌胃相应中药,连续14 d。用酶联免疫法(ELISA)测定外周血和肺泡灌洗液中细胞因子IL-8、IL-1β、TNF-α和IL-10的含量。结果①与正常组比较,模型组大鼠外周血和肺泡灌洗液的IL-8、IL-1β、TNF-α和IL-10含量均升高(P<0.01或P<0.05)。②与模型组比较,治肠组外周血和肺泡灌洗液的IL-8、IL-1β、TNF-α和IL-10含量均降低(P<0.01或P<0.05)。③与治肺组比较,肺肠同治组外周血IL-1β含量和肺泡灌洗液IL-8含量降低(P<0.01或P<0.05)。结论通利大肠或在治肺的基础上增加通利大肠,能降低COPD模型大鼠的外周血和肺泡灌洗液中的细胞因子含量,从而减轻全身及局部的炎症反应,这可能是COPD“从肠论治”效应产生的作用环节之一。
Objective To observe the effect of Tonglida Decoction on the expression of interleukin-8 (IL-8), interleukin-1β (IL-1β), tumor necrosis factor α (TNF-α) and interleukin-10 (IL-10) levels in patients with chronic obstructive pulmonary disease (COPD). Methods COPD rat models were established by tracheal injection of lipopolysaccharide plus cigarette smoking, and were randomly divided into normal group, model group, treatment group and treatment group. Normal group, model group, gavage pure water, each administration group gavage the corresponding Chinese medicine, continuous 14 d. The contents of cytokines IL-8, IL-1β, TNF-α and IL-10 in peripheral blood and BALF were determined by enzyme-linked immunosorbent assay (ELISA) Results ① Compared with normal group, the levels of IL-8, IL-1β, TNF-α and IL-10 in peripheral blood and BALF in model group were significantly increased (P <0.01 or P <0.05). ②Compared with model group, the levels of IL-8, IL-1β, TNF-α and IL-10 in peripheral blood and alveolar lavage fluid in treatment group were decreased (P <0.01 or P <0.05). ③Compared with the control lung group, the content of IL-1β in peripheral blood and the content of IL-8 in the bronchoalveolar lavage fluid decreased (P <0.01 or P <0.05). Conclusion Tongli intestine or Tongliao large intestine in the lungs to increase, can reduce the COPD model of rat peripheral blood and alveolar lavage fluid cytokine content, thereby reducing the systemic and local inflammatory response, which may be COPD “From the rule of law,” the effect of the effect of one of the links.
