目的分析胰岛素样生长因子Ⅱ(insulin-like growth factorⅡ,IGF-Ⅱ)及胰岛素样生长因子ⅡmRNA结合蛋白3(insulin-like growth factorⅡmRNA binding protein 3,IMP3)在宫颈组织中的表达及意义。方法收集2013年1月至2015年3月德州市人民医院妇科就诊患者取活检或者手术切除后的宫颈组织标本85例,其中宫颈癌42例,正常宫颈组织15例,宫颈炎10例,宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)Ⅰ期7例、Ⅱ-Ⅲ期11例,采用免疫组化的方法检测组织中IGF-Ⅱ及IMP3蛋白表达情况,对不同宫颈组织标本的IGF-Ⅱ及IMP3蛋白阳性表达率进行比较,并分析IGF-Ⅱ及IMP3蛋白的表达与宫颈癌临床病理特征之间的关系。结果不同宫颈组织标本的IGF-Ⅱ蛋白阳性表达率不全相等(P<0.05):CINⅡ-Ⅲ期组及宫颈癌组的IGF-Ⅱ蛋白阳性表达率高于正常组,宫颈癌组的IGF-Ⅱ蛋白阳性表达率高于宫颈炎组、CINⅠ期组及CINⅡ-Ⅲ期组(P<0.05)。不同宫颈组织标本的IMP3蛋白阳性表达率不全相等(P<0.05):宫颈癌组的IMP3蛋白阳性表达率高于正常组、宫颈炎组、CINⅠ期组及CINⅡ-Ⅲ期组(P<0.05)。不同年龄、人乳头瘤病毒(human papillomavirus,HPV)感染与否、肿瘤分化程度及不同病理类型之间患者的IGF-Ⅱ及IMP3蛋白的阳性表达率比较差异无统计学意义(P>0.05)。结论 IGF-Ⅱ及IMP3蛋白表达可为宫颈癌诊断提供一定的依据,可将其作为宫颈癌筛查的生物学标志物之一。
Objective To analyze the expression and significance of insulin-like growth factor Ⅱ (IGF-Ⅱ) and insulin-like growth factor Ⅱ mRNA (IMP3) in cervical tissue. Methods Totally 85 cases of cervical tissue collected from patients with gynecological examination in Dezhou People’s Hospital from January 2013 to March 2015 were collected from 42 cases of cervical cancer, 15 cases of normal cervical tissue, 10 cases of cervicitis, Seven cases of cervical intraepithelial neoplasia (CIN) stage Ⅰ and 11 cases of stage Ⅱ-Ⅲ immunohistochemistry were used to detect the expression of IGF-Ⅱand IMP3 protein in different tissues of the cervix, IMP3 protein expression were compared and analyzed the relationship between the expression of IGF-Ⅱ and IMP3 protein and clinicopathological features of cervical cancer. Results The positive rates of IGF-Ⅱprotein expression in different cervical tissues were not equal (P <0.05). The positive rates of IGF-Ⅱprotein in CINⅡ-Ⅲgroup and cervical cancer group were higher than those in normal group and cervical cancer group The protein positive expression rate was higher than cervicitis group, CINⅠstage group and CINⅡ-Ⅲstage group (P <0.05). The positive rate of IMP3 protein in cervical cancer group was higher than that in normal group, cervicitis group, CINⅠstage group and CINⅡ-Ⅲstage group (P <0.05) . There was no significant difference in the positive expression rates of IGF-Ⅱand IMP3 between patients with different age, human papillomavirus (HPV) infection, tumor differentiation and different pathological types (P> 0.05). Conclusion The expression of IGF-Ⅱ and IMP3 may provide a basis for the diagnosis of cervical cancer, which can be used as one of the biomarkers of cervical cancer screening.
