目的:了解宫颈鳞状上皮癌化疗前后BCL-2的变化。方法:分别给予宫颈癌HeLa細胞不同浓度GA处理24 h,用annexinV方法检测细胞凋亡,RT-PCR检测目的基因的表达,用GA处理HeLa細胞4、16、24、48 h后,用免疫印记分析相关Bcl-2蛋白水平表达的变化。结果:宫颈癌HeLa細胞经不同浓度的GA作用24 h,Bcl-2蛋白表达水平呈时间依赖性下降,Bcl-2 mRNA表达呈剂量依赖性下降,细胞凋亡指数呈剂量依赖性增强,处理组与对照组之间的差异有统计学意义。结论:宫颈癌HeLa細胞抗凋亡基因Bcl-2的转录和表达,促进凋亡。
Objective: To investigate the changes of BCL-2 in cervical squamous carcinoma before and after chemotherapy. Methods: Human cervical cancer HeLa cells were treated with different concentrations of GA for 24 hours. The apoptosis of HeLa cells was detected by annexinV method. The expression of target gene was detected by RT-PCR. After being treated with GA for 4, 16, 24 and 48 hours, Changes in the expression of related Bcl-2 protein levels were analyzed. Results: The cervical cancer HeLa cells treated with different concentrations of GA for 24 h, the expression of Bcl-2 protein decreased in a time-dependent manner, the expression of Bcl-2 mRNA decreased in a dose-dependent manner, and the apoptotic index increased in a dose-dependent manner. The difference between the control group and the control group was statistically significant. Conclusion: The transcription and expression of anti-apoptotic gene Bcl-2 in cervical cancer HeLa cells promote apoptosis.
