目的通过研究缩泉丸对肾虚多尿大鼠垂体ACTH mRNA及蛋白表达的影响,阐明其补肾缩尿的机制。方法采用腺嘌呤造成大鼠肾虚多尿模型,利用酶联免疫分析法分别检测正常对照组、模型对照组、缩泉丸(高、中、低剂量)组、金锁固精丸组动物血中ACTH、ALD的含量;并采用RT-PCR、免疫组化的方法检测各组动物垂体ACTH mRNA及蛋白的表达。结果与模型对照组比较,缩泉丸中、高剂量组可显著提高肾虚多尿大鼠血中ACTH、ALD含量(P<0.05~0.01);缩泉丸高剂量组可使肾虚多尿大鼠垂体ACTH mRNA表达明显增高(P<0.01);缩泉丸低中高剂量组均可使肾虚多尿大鼠垂体ACTH蛋白阳性产物的表达增强,表达区域广泛分布于前叶和中间叶。结论缩泉丸可上调肾虚多尿大鼠垂体ACTH mRNA及蛋白的表达,增强ACTH的功能,从而促进ALD的合成,发挥调节水液代谢的作用。
Objective To study the influence of shrink spring pill on the expression of ACTH mRNA and protein in pituitary of rats with kidney-yang polyuria and to elucidate the mechanism of kidney invigorating and urination. Methods Adenine was used to induce kidney-derived polyuria in rats. Serum levels of the normal control group, model control group, shrink-spring pill (high, middle and low dose) ACTH and ALD were detected. The expressions of ACTH mRNA and protein in pituitary of each group were detected by RT-PCR and immunohistochemistry. Results Compared with the model control group, Tongquan pill medium and high dose groups could significantly increase the blood ACTH and ALD levels (P <0.05 ~ 0.01) Pituitary ACTH mRNA expression was significantly increased (P <0.01); shrink Quan Wan low medium and high dose groups can make kidney pituitary ACTH protein expression increased pituitary, the expression region is widely distributed in the anterior lobe and middle lobe. Conclusions Shuxuquan Pill can upregulate the expression of ACTH mRNA and protein in the pituitary of rats with kidney-deficiency and polyuria, enhance the function of ACTH, promote the synthesis of ALD, and play an important role in the regulation of water-fluid metabolism.
