目的:探讨钩吻醇提-氯仿萃取(GW-B2)组分对Hela细胞生长与增殖以及细胞周期的影响。方法:采用XTT法分析细胞生长与增殖,Timelapse显微镜观察细胞生长和形态,流式细胞仪检测凋亡和细胞周期,综合评价GW-B2组分抗Hela细胞的作用。结果:GW-B2具有明显抑制Hela细胞生长与增殖的作用,1μL/mL GW-B2组分处理的Hela细胞生长速度明显降低,在2~5μL/mL的剂量范围内,细胞生长与增殖能力基本完全被抑制。GW-B2组分处理后,Hela细胞主要的形态学变化为贴壁能力丧失,悬浮细胞增多,细胞分裂阻滞,部分细胞崩解破碎。GW-B2组分能明显的诱导Hela细胞凋亡,1μL/mL、2μL/mL和5μL/mL GW-B2组分处理72h后,Hela细胞的凋亡率分别可达8.16%、36.83和61.32%,而对细胞周期的影响主要是G2/M期阻滞,同时对G0/G1期也有一定的阻滞作用。结论:GW-B2组分具有抑制Hela细胞增殖、诱导凋亡,具有明显的抗肿瘤细胞的作用;GW-B2组分对细胞周期的影响主要是G2/M期阻滞,随后可诱导细胞的凋亡。
Objective: To investigate the effects of ghrelin-chloroform extraction (GW-B2) on the growth, proliferation and cell cycle of Hela cells. METHODS: Cell growth and proliferation were analyzed by XTT method. Cell growth and morphology were observed by Timelapse microscopy. Apoptosis and cell cycle were measured by flow cytometry. The effects of GW-B2 components against Hela cells were comprehensively evaluated. RESULTS: GW-B2 significantly inhibited the growth and proliferation of Hela cells. The growth rate of HeLa cells treated with 1 μL/mL GW-B2 was significantly decreased. The growth and proliferation of Hela cells were basically in the dose range of 2-5 μL/mL. Completely suppressed. After GW-B2 treatment, the main morphological changes of Hela cells were the loss of adherent capacity, the increase of suspended cells, the arrest of cell division, and the breakdown of some cells. GW-B2 could obviously induce the apoptosis of Hela cells. After treatment with GW-B2 at 1μL/mL, 2μL/mL and 5μL/mL for 72h, the apoptosis rate of Hela cells reached 8.16%, 36.83% and 61.32%, respectively. The effect on the cell cycle is mainly the G2/M phase block, and it also has a certain blocking effect on the G0/G1 phase. Conclusion: GW-B2 can inhibit the proliferation of Hela cells and induce apoptosis. It has obvious effect on anti-tumor cells. The effect of GW-B2 on the cell cycle is mainly G2/M arrest, and then the cells can be induced. Apoptosis.
