目的探讨冬凌草甲素(oridonin,Ori)对人乳腺癌细胞MCF-7的增殖、周期及凋亡的影响。方法以不同浓度的Ori处理MCF-7细胞,MTT法检测细胞生长抑制率;应用流式细胞分析术检测细胞周期和凋亡。结果 20~50μM的Ori可显著抑制MCF-7细胞的增殖(P<0.01),50μMOri作用24、36h后细胞生长抑制率分别为67.37%和97.63%。50μmOri处理24h后,MCF-7细胞的凋亡率为80.03%。结论冬凌草甲素可显著抑制MCF-7细胞的生长,诱导细胞周期阻滞和细胞凋亡,是一种潜在的抗乳腺癌药物。
Objective To investigate the effects of oridonin (Ori) on the proliferation, cell cycle and apoptosis of human breast cancer cell line MCF-7. Methods MCF-7 cells were treated with different concentrations of Ori. Cell growth inhibition rate was measured by MTT assay. Cell cycle and apoptosis were detected by flow cytometry. Results Ori inhibited the proliferation of MCF-7 cells at a dose of 20-50 μM (P <0.01). The inhibitory rates of cell growth were 67.37% and 97.63% after treated with 50 μM Ori for 24 and 36 h, respectively. After treated with 50μmOri for 24 hours, the apoptosis rate of MCF-7 cells was 80.03%. Conclusion Oridonin can significantly inhibit the growth of MCF-7 cells, induce cell cycle arrest and apoptosis, and is a potential anti-breast cancer drug.
