采用C18反相柱层析法分离大豆异黄酮糖苷,再将其水解制备成异黄酮苷元,研究其对结肠癌HT-29和肺癌A549细胞增殖和凋亡的影响。用MTT比色法观察大豆异黄酮对肿瘤细胞增殖的影响,用流式细胞术检测大豆异黄酮对细胞周期分布的影响,用TUNEL染色法检测其对HT-29细胞凋亡的影响。结果表明,大豆异黄酮呈时间和浓度依赖性地抑制肿瘤细胞增殖,诱导肿瘤细胞凋亡。同时改变结肠癌HT-29细胞周期分布,使其阻滞于G2/M期。提示,大豆异黄酮可通过改变细胞周期分布和诱导细胞凋亡发挥抗肿瘤作用。
Soy isoflavone glucoside was isolated by C18 reverse phase column chromatography and then hydrolyzed to prepare isoflavone aglycone to study the effect on proliferation and apoptosis of colon cancer HT-29 and lung cancer A549 cells. The effect of soybean isoflavones on the proliferation of tumor cells was observed by MTT colorimetric assay. The effect of soybean isoflavones on cell cycle distribution was detected by flow cytometry. The apoptosis of HT-29 cells was detected by TUNEL staining. The results showed that soy isoflavones inhibited the proliferation of tumor cells in a time and concentration-dependent manner, and induced the apoptosis of tumor cells. At the same time, the cell cycle distribution of HT-29 colon cancer was changed to arrest in G2 / M phase. Tip, soy isoflavones can change the cell cycle distribution and induce apoptosis of anti-tumor effect.
