目的比较纳米增强免疫比浊法(NETIA)与高效液相色谱法(HPLC)检测化疗患者5-氟尿嘧啶(5-FU)血药浓度结果。方法肠癌或鼻咽癌患者45例接受包含连续输注5-FU的治疗方案。分别采用NETIA和HPLC检测5-FU血药浓度。结果 NETIA与HPLC检测5-FU的平均回收率、精密度、准确度和线性均较理想。NETIA和HPLC检测的5-FU血药浓度分别为82.3~1181.0ng/ml和99.0~1091.4ng/ml。Spearman相关性分析结果显示,两种方法检测的5-FU血药浓度呈正相关(r_s=0.751,P<0.01)。结论 NETIA与HPLC检测5-FU血药浓度的相关性较好。与HPLC比较,NETIA检测5-FU血药浓度操作简单、快速,可用于临床5-FU血药浓度监测。
Objective To compare the plasma concentrations of 5-fluorouracil (5-FU) in patients undergoing chemotherapy with neutrophil nephelometry (NETIA) and high performance liquid chromatography (HPLC) Methods Forty-five patients with colorectal cancer or nasopharyngeal carcinoma received a regimen that included continuous infusion of 5-FU. NETIA and HPLC were used to detect 5-FU blood concentration. Results The average recovery, precision, accuracy and linearity of 5-FU detected by NETIA and HPLC were all satisfactory. The 5-FU plasma concentrations of NETIA and HPLC were 82.3 ~ 1181.0ng / ml and 99.0 ~ 1091.4ng / ml, respectively. Spearman correlation analysis showed that there was a positive correlation between serum 5-FU concentrations detected by the two methods (r_s = 0.751, P <0.01). Conclusion The correlation between NETIA and HPLC in detecting the concentration of 5-FU is good. Compared with HPLC, NETIA detection of 5-FU plasma concentration is simple, rapid, and can be used for clinical 5-FU plasma concentration monitoring.
