目的探讨CD45分子在γδT细胞发育过程中的作用。方法采用CD45基因敲除变种鼠,观察其Vγ3树突状表皮T细胞(Vγ3DETC),这类唯一居住于鼠类表皮组织的γδT细胞的变化。以原位免疫标记观察其表皮内Vγ3DETC的分布密度和免疫表型,以RT-PCR检测表皮细胞中Vγ3TCRmRMA的表达水平,以双荧光FCM测定Vγ3DETC在表皮细胞中及Vγ2T细胞在淋巴结细胞中所占比例,并与野生型鼠作对照。结果CD45缺失鼠表皮内Vγ3DETC的密度、免疫表型,表皮细胞Vγ3TCRmRNA表达水平以及Vγ3DETC和Vγ2T细胞分别在表皮细胞及淋巴结细胞中的比例与野生型鼠无明显差异。结论CD45分子虽在αβT细胞发育中起重要作用,但对包括Vγ3DETC及Vγ2T细胞在内的γδT细胞之成熟过程可能并非必需
Objective To investigate the role of CD45 in the development of γδT cells. Methods CD45 knockout mice were used to observe the changes of Vγ3 dendritic cells (Vγ3DETC), the only γδT cells in mouse epidermis. The distribution density and immunophenotype of Vγ3DETC in epidermis were observed by in situ immunolabeling. The expression level of Vγ3TCRmRMA in epidermal cells was detected by RT-PCR, and the expression of Vγ3DETC in epidermal cells and Vγ2T cells in lymph node cells by dual-fluorescence FCM Ratio, and compared with wild-type mice. Results The density, immunophenotype, Vγ3TCRmRNA expression level of Vγ3DETC and the ratio of Vγ3DETC and Vγ2T cells in epidermal cells and lymph node cells in CD45 null mice were not significantly different from those in wild type mice. Conclusions Although the CD45 molecule plays an important role in the development of αβT cells, the maturation of γδT cells including Vγ3DETC and Vγ2T cells may not be necessary
