目的 探讨高渗醋酸钠 右旋糖酐 (HA D)对大鼠创伤失血性休克后肝细胞线粒体功能的影响。方法 大鼠 6 4只 ,随机分为正常对照组、休克末组、生理盐水 (NS ,4ml·kg- 1 体重 )治疗后 30、6 0、12 0min组及HAD(4ml·kg- 1 体重 )治疗后 30、6 0、12 0min组 ,每组 8只。股骨骨折加股动脉放血制作创伤失血性休克模型 ,各组动物至实验时限后活杀 ,取肝组织测定肝细胞线粒体呼吸控制率 (RCR)、腺苷二磷酸 氧 (AdenosinediphosphateADP O)、线粒体钙含量、线粒体内膜H+ ATP酶及辅酶Ⅰ 辅酶Q(NADH Co .Q)还原酶活性。结果 复苏后 6 0和 12 0min ,HAD组的RCR、ADP O、H+ ATP酶及NADH Co .Q还原酶活性均非常显著地高于NS组 ,线粒体钙含量则非常显著地低于NS组 (P <0 .0 1)。结论 小剂量HAD复苏能显著减轻创伤失血性休克大鼠肝细胞线粒体功能的损伤
Objective To investigate the effect of hypertonic sodium dextran (HA D) on the mitochondrial function of hepatocytes after traumatic hemorrhagic shock in rats. Methods Sixty-four rats were randomly divided into normal control group, shock end group, saline group (NS, 4ml · kg-1 body weight), 30,6 0,12 0min group and HAD (4ml · kg-1 body weight) After treatment, 30,6 0,12 0 min group, each group of 8. Femoral fractures and femoral artery bleeding were used to make the model of traumatic hemorrhagic shock. Animals in each group were sacrificed after the experimental period, and the liver tissue was taken to measure mitochondrial respiratory control rate (RCR), adenosine diphosphate (ADP O), mitochondrial calcium , Mitochondrial inner membrane H + ATPase and coenzyme Ⅰ coenzyme Q (NADH Co. Q) reductase activity. Results At 60 and 120 min after resuscitation, the activities of RCR, ADP O, H + ATPase and NADH Co. Q reductase in HAD group were significantly higher than those in NS group (P <0.05). The mitochondrial calcium content in HAD group was significantly lower than that in NS group <0 .0 1). Conclusion Low dose HAD resuscitation can significantly reduce the damage of mitochondrial function of hepatocytes in traumatic hemorrhagic shock rats
