目的:探究胃癌的化疗联合免疫细胞治疗过程中树突状细胞(DC)和自然杀伤(NK)细胞之间Cross-talk 的机制。方法选取2014年1~12月河南科技大学第一附属医院新区医院收治的需进行细胞治疗的胃癌患者80例,将其随机分为试验组和对照组,各40例。对照组在化疗前1周实施DC 疫苗联合NK 细胞方案;试验组,化疗后1周实施肿瘤DC 联合NK 细胞的治疗方案。共设置4个测量时间点:治疗前24 h、治疗后48 h、治疗后1周、治疗后4周。使用酶联免疫吸附实验法监测患者血清中可溶性细胞因子的表达,包括白介素-12、白介素-18、白介素-15、分泌型肿瘤坏死因子-α(TNF-α)以及分泌型干扰素(IFN-C)。结果试验组和对照组血清中的白介素-12、白介素-18和白介素-15在治疗前差异均无统计学意义(P>0.05)。治疗后48 h、治疗后1周、治疗后4周时,试验组患者血清中DC 和NK 细胞相关可溶性细胞因子的表达较对照组数据明显增高(均P<0.05),差异均有统计学意义。其表达在治疗后1周达到峰值,其峰值维持治疗后4周(P<0.01)。结论在化疗治疗之后进行免疫细胞治疗,免疫细胞间的对话(Cross-talk)调控诱发的DC 和NK 细胞间的正反馈作用显著增强。“,”ObjectiveStudy on the regulation mechanism of Cross-talking of NK and DC in gastric cancer by chemotherapy combined with immunotherapy.Methods Using repeated measure design. Patients with gastric cancer (of either sex) for cell therapy in 80 patients.The 80 patients were randomly divided into 2 groups (n=40):In the control group,the tumor DC vaccine combined with NK cells was implemented in the previous week.In the experimental group,the tumor DC vaccine combined with NK cells was carried out one week after chemotherapy.Set a total of 4 time points:before treatment,after treatment 24 h;48 h;after treatment of 1week and 4week after treatment.ELISA method was used to monitor the expression of soluble cytokines in serum,including IL-12,IL-18,IL-15,TNF- and IFN-C.Results There was no difference in IL-15,IL-18 and IL-12 between the experimental group and the control group (P<0.05). After treatment, the serum levels of DC cells in the experimental group were significantly higher than that of the control group,IL-12 (P<0.05),IL-18 (P<0.05),IL-15 (P<0.05),and the expression of 1week reached the peak after treatment,and the peak value of 4week (P<0.05) was maintained.Conclusions After chemotherapy treatment,the positive feedback between DC cells and NK cells induced by Cross-talk was significantly enhanced by immune cell therapy.
