目的:(1)研究喉癌多药耐药细胞株的耐药谱,为实现临床化学治疗个体化提供实验依据。(2)研究维拉帕米对喉癌细胞株耐药逆转浓度及耐药逆转谱。方法:以喉癌KBV200耐药细胞株为研究对象:(1)采用MTT法检测临床常用8种抗肿瘤药物对该细胞株的增殖抑制作用。(2)采用MTT法检测维拉帕米对喉癌细胞株耐药逆转作用。结果:(1)长春新碱、阿霉素、平阳霉素、环磷酰胺、阿糖胞苷对KBV200耐药细胞株的抑制作用差,三尖杉酯、5-Fu及顺铂对KBV200耐药细胞株的抑制作用好。(2)维拉帕米(12.5μg/ml)能逆转长春新碱、阿霉素、平阳霉素、环磷酰胺对KBV200耐药细胞株的耐药作用。结论:(1)KBV200耐药细胞株对长春新碱、阿霉素、平阳霉素、环磷酰胺、阿糖胞苷5种药物有不同程度耐药。(2)维拉帕米(12.5μg/ml)对KBV200耐药细胞株的耐药逆转谱是长春新碱、阿霉素、平阳霉素、环磷酰胺。
Objectives: (1) To study the drug resistance spectrum of multidrug-resistant laryngeal carcinoma cell lines and provide experimental evidence for individualization of clinical chemotherapy. (2) to study the verapamil drug-resistant laryngeal cancer cell line reversal concentration and drug resistance reversal spectrum. Methods: The KBV200 cell line was used as research object. (1) MTT assay was used to detect the inhibitory effect of 8 anti-tumor drugs commonly used in clinic on the proliferation of this cell line. (2) MTT assay was used to detect the reversal effect of verapamil on the laryngeal carcinoma cell line. Results: (1) Vincristine, doxorubicin, pingyangmycin, cyclophosphamide, cytarabine inhibition of KBV200 drug-resistant cell line was poor, and the effect of harringtonine, 5-Fu and cisplatin on KBV200 Good inhibition of drug-resistant cell lines. (2) Verapamil (12.5μg / ml) could reverse the drug resistance of vincristine, doxorubicin, pingyangmycin and cyclophosphamide to KBV200 resistant cell line. Conclusion: (1) KBV200 resistant cell lines have different degrees of resistance to vincristine, doxorubicin, pingyangmycin, cyclophosphamide and cytarabine. (2) The reversal spectrum of drug resistance of verapamil (12.5μg / ml) to KBV200 cell line was vincristine, doxorubicin, bleomycin and cyclophosphamide.
