目的:探讨子宫骶韧带局部组织中同源盒基因A11(HOXA11)、雌激素受体α(ER-α)、雌激素受体β(ER-β)的表达及胶原纤维、弹力纤维形态的改变与盆腔器官脱垂(POP)发生的关系。方法:采用免疫组织化学PV法检测24例(POP及非POP各12例,分别为研究组和对照组)子宫骶韧带组织中HOXA11、ER-α、ER-β的表达,应用间苯二酚品红法和丽春红酸性品红法分别对弹性纤维和胶原纤维进行染色。结果:①HE染色显示研究组子宫骶韧带组织有胞浆水肿、空泡变性及淀粉样变等不同程度的变性。②与对照组相比,研究组胶原纤维染色较浅,弹力纤维弯曲度和密度减低,纤维碎裂、崩解或聚集成团块状,排列相对不规则。③研究组HOXA11、ER-α和ER-β表达均减弱,与对照组相比,差异有统计学意义(P<0.05)。结论:POP患者子宫骶韧带组织发生变性,其弹力纤维的形态遭到破坏,胶原纤维的含量减少。HOXA11、ER-α和ER-β在POP患者子宫骶韧带组织中的表达均降低,提示均参与POP的发生。
Objective: To investigate the expression of HOXA11, ER-α and ER-β in the local tissues of the uterosacral ligaments and the morphological changes of collagen fibers and elastic fibers And pelvic organ prolapse (POP) the relationship between the occurrence. Methods: The expression of HOXA11, ER-α and ER-β in uterosacral ligaments of 24 cases (POP and non-POP in 12 cases, study group and control group respectively) was detected by immunohistochemistry PV method. The magenta and Ponceau acid magenta methods stain elastic and collagen fibers, respectively. Results: ①HE staining showed that the study group uterosacral ligament tissue cytoplasm edema, vacuolar degeneration and amyloidosis, such as varying degrees of degeneration. ②Compared with the control group, the collagen fibers in the study group were lighter in color, the curvature and density of the elastic fibers were reduced, and the fibers were disintegrated or disintegrated or aggregated into a clumpy shape with relatively irregular arrangement. ③ The expression of HOXA11, ER-α and ER-βwere weakened in the study group compared with the control group, the difference was statistically significant (P <0.05). Conclusion: The uterosacral ligament tissues of POP patients degenerate, the morphology of the elastic fibers is destroyed and the content of collagen fibers is reduced. The expression of HOXA11, ER-α and ER-β in the uterosacral ligament tissue of POP patients were decreased, suggesting that all involved in the occurrence of POP.
