生脉散对阿霉素致心力衰竭大鼠的症状改善作用研究

目的探讨生脉散水煎液对阿霉素致心力衰竭(心衰)大鼠的症状改善作用。方法 2014年5—12月采用阿霉素建立大鼠心衰模型,模型建立成功后随机分为正常组、模型组、地高辛组、生脉散组各10只。地高辛组给予0.3 mg/kg地高辛灌胃给药,生脉散组给予按原方比例折算的生脉散煎剂灌胃,正常组及模型组均给予等量蒸馏水灌胃。给药四周后,检测各组大鼠心功能指标即左心室舒张末内径(left ventricular end-diastolic dimension,LVEDD)、左心室收缩末内径(left ventricular end-systolic dimension,LVESD)水平,并用ELISA法检测大鼠血液中脑钠肽(brain natriuretic peptide,BNP)、心肌肌钙蛋白I(cardiac troponin I,c Tn I)、血管紧张素Ⅱ(AngiotensinⅡ,AngⅡ)、醛固酮(Aldosterone,ALD)的水平,同时检测白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的水平。计量资料采用方差分析,两两比较采用LSD-t检验,P<0.05为差异有统计学意义。结果给药4周后,与模型组相比,生脉散组大鼠体重显著升高,血液中BNP、c Tn I、AngⅡ和ALD水平显著下降,LVEDD、LVESD水平显著下降,IL-6、TNF-α水平显著下降,差异均有统计学意义(均P<0.05)。结论生脉散水煎液能够显著改善心衰大鼠的症状,且用药安全。 Objective To investigate the effect of Shengmai San decoction on symptoms of doxorubicin-induced heart failure (CHF) in rats. Methods The rat model of heart failure was established by adriamycin from May to December in 2014. The model was established and randomly divided into normal group, model group, digoxin group and Shengmai Powder group. Digoxin group was given 0.3 mg / kg digoxin intragastric administration, Shengmaisan group given proportion of the original conversion of Shengmai decoction gavage, normal group and model group were given the same amount of distilled water gavage. Four weeks after the administration, the left ventricular end-diastolic dimension (LVEDD) and left ventricular end-systolic dimension (LVESD) were measured in each group. The levels of brain natriuretic peptide (BNP), cardiac troponin I (cTn I), Angiotensin II (Ang II) and aldosterone (ALD) The levels of interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) were measured simultaneously. Measurement data were analyzed by ANOVA, pairwise comparisons using LSD-t test, P <0.05 for the difference was statistically significant. Results Compared with the model group, the body weight of rats in Shengmai powder group increased significantly after 4 weeks of administration. The levels of BNP, cTn I, Ang II and ALD were significantly decreased, while the levels of LVEDD and LVESD were significantly decreased. The levels of IL-6, TNF-α levels were significantly decreased, the differences were statistically significant (P <0.05). Conclusion Shengmai San decoction can significantly improve the symptoms of heart failure rats, and medication safety.
