以西藏卡门地区工布乌头叶片为材料,研究影响其DNA提取效果的关键因子,以期建立最优提取体系。该研究采用传统CTAB法、改良CTAB法(增加了2%的PVP、β-巯基乙醇)、SDS法、改良SDS法(异丙醇进行-20℃预冷)、试剂盒法(吸附柱)、改良试剂盒法6种方法对工布乌头叶片DNA提取并进行比对,结果表明,采用改良后的方法,提取的DNA质量较好,且改良CTAB法提取效果更佳;其中改良试剂盒法提取的DNA较纯,琼脂糖凝胶电泳条带最亮,杂质少;若考虑经济因素,则以改良CTAB法为优。
In order to establish the optimal extraction system, the key factors affecting the DNA extraction efficiency were studied in the area of Carmen head in Camellia, Tibet. In this study, the traditional CTAB method, modified CTAB method (2% increase of PVP and β-mercaptoethanol), SDS method, modified SDS method (isopropanol precooling at -20 ℃), kit method Improved kit method six kinds of methods to extract and compare the DNA of Aconitum mongolicum leaves, the results showed that using improved method, the quality of the extracted DNA is better, and the improved CTAB method is better; the improved kit method DNA extracted more pure, agarose gel electrophoresis strip the brightest, less impurities; if economic factors, the improved CTAB method is excellent.
