以相应引物对凡纳滨对虾(Litopenaeus vannamei)线粒体DNA细胞色素氧化酶I亚基基因(mtDNA CO I)进行PCR扩增,经过基因重组、转化、克隆、筛选、DNA测序,得到709 bp的碱基片段.碱基组成A、C、G、T含量分别为198 bp(27.93%)、136 bp(19.18%)、129 bp(18.19%)2、46 bp(34.70%).与果蝇(Drosophila yakuba)的mtDNA CO I全序列比对,经分析发现:本实验获得的凡纳滨对虾mtCO I基因片段序列和GenBank上的同种序列(AY264901)都只是基因序列的一部分,二者之间有41 bp的重叠并可拼接,拼接后的总长为1 515 bp.证明本实验条件下获得的mtCO I基因片段确实来自凡纳滨对虾线粒体DNA,且本实验所用引物具有通用性.
