目的研究表皮生长因子对人牙周成纤维细胞增殖的作用。方法原代培养人牙周成纤维细胞,MTT法检测表皮生长因子作用后牙周成纤维细胞的增殖情况,ELISA法检测细胞培养上清中细胞周期蛋白cyclin D1的表达。结果各浓度表皮生长因子均可以促进牙周成纤维细胞的生长,与正常对照组比较,36 h 20 ng/ml组、72 h 10和20 ng/ml组有统计学差异(P<0.05);ELISA检测发现细胞培养上清中cyclin D1的表达增加,20 ng/ml组有统计学差异(P<0.05)。结论表皮生长因子通过上调细胞周期蛋白cyclin D1的表达而促进牙周成纤维细胞的增殖。
Objective To study the effect of epidermal growth factor on the proliferation of human periodontal fibroblasts. Methods Human periodontal fibroblasts were cultured in primary culture. The proliferation of periodontal fibroblasts was detected by MTT assay. The expression of cyclin D1 in cell culture supernatants was detected by ELISA. Results Epidermal growth factor (EGF) at various concentrations could promote the growth of periodontal fibroblasts. Compared with the normal control group, the levels of 36 h 20 ng / ml, 72 h 10 and 20 ng / ml groups were significantly different (P <0.05). The result of ELISA showed that the expression of cyclin D1 increased in the cell culture supernatant, and there was a significant difference (P <0.05) in 20 ng / ml group. Conclusion Epidermal growth factor promotes the proliferation of periodontal fibroblasts by up-regulating the expression of cyclin D1.
