目的:探讨早孕因子(EPF)活性物对小鼠脾淋巴细胞增殖的影响及其在胎母免疫中的作用。方法:使用离子交换层析、亲和层析和凝胶过滤技术提纯3~6周孕妇血清中EPF;用淋巴细胞转化法观察EPF对淋巴细胞的作用。结果:EPF活性物G4、G3及孕血清均能抑制淋巴细胞DNA合成,尤以G4作用更为明显,并呈一定剂量依赖性。结论:EPF可能在介导、调控妊娠早期诱导的免疫抑制,保证妊娠的持续中起重要作用
Objective: To investigate the effect of early pregnancy factor (EPF) on the proliferation of mouse splenic lymphocytes and its role in the immune of the fetus. Methods: EPF was purified from serum of 3 ~ 6 weeks pregnant women by ion exchange chromatography, affinity chromatography and gel filtration. The effect of EPF on lymphocytes was observed by lymphocyte transformation. Results: The EPF actives G4, G3 and serum of pregnant women could inhibit the DNA synthesis of lymphocytes, especially the effect of G4 was more obvious and in a dose-dependent manner. Conclusion: EPF may play an important role in mediating and regulating the immunosuppression induced during early pregnancy and ensuring the continuation of pregnancy
