目的:建立高效毛细管电泳法测定伪麻黄碱的人体血药浓度。方法:采用石英毛细管柱(45 cm×50μm),运行电压20 kV,温度25℃,30 mB(3.15 Pa)压力进样,时间15 s;缓冲液为0.05 mol·L-1硼砂溶液;检测波长为210 nm。结果:线性范围50~500 ng·ml-1(r=0.999 6),平均回收率79.20%,日内、日间RSD<5%,最低定量浓度为25 ng·ml-1。结论:该法简便、快速、准确,适用于伪麻黄碱药物动力学和生物等效性研究。
Objective: To establish a high performance capillary electrophoresis method for the determination of pseudoephedrine in human plasma concentration. METHODS: The sample was injected with quartz capillary column (45 cm × 50 μm), operating voltage of 20 kV, temperature of 25 ℃ and pressure of 30 mB (3.15 Pa) for 15 s. The buffer was 0.05 mol·L -1 borax Solution; detection wavelength of 210 nm. Results: The linear range was 50-500 ng · ml-1 (r = 0.999 6). The average recovery was 79.20%. The intra-day and inter-day RSD was less than 5% and the lowest concentration was 25 ng · ml-1. Conclusion: The method is simple, rapid and accurate and suitable for the study of pharmacokinetics and bioequivalence of pseudoephedrine.
