对工程蝇蛆体内活性物质及体外抗肿瘤作用进行了研究。选用SephadexG-50初步分离蝇蛆体内活性物质;采用MTT法测定K562细胞的生长抑制率;通过倒置显微镜观察K562细胞的形态变化。实验结果表明:峰1组分半致死浓度IC50为100μg/mL,峰2组分半致死浓度IC50为80μg/mL。随所添加活性物质浓度的增大,肿瘤细胞受抑制作用增强;活性物质作用于K562细胞前后其细胞形态发生明显变化,受抑制细胞由一个完整细胞变为多个凋亡小体,因此可以说明工程蝇蛆体内活性物质在体外对K562细胞具有抑制生长的作用。
The active substances in the fly maggot and the anti-tumor effect in vitro were studied. Sephadex G-50 was used to separate the active substances in the maggot; the growth inhibition rate of K562 cells was determined by MTT assay; the morphological changes of K562 cells were observed by inverted microscope. The experimental results showed that the IC50 of peak 1 was 100μg / mL, and the IC50 of peak 2 was 80μg / mL. With the increase of the concentration of the active substance added, the inhibition of the tumor cells was enhanced. The active substance had a significant change in morphology before and after K562 cells, and the inhibited cells changed from an intact cell into many apoptotic bodies. Therefore, Muskrat body active substances in vitro inhibit the growth of K562 cells.
