多年来的研究结果对肝素稳定性为pH的函数关系颇有争议。有人认为,在5%葡萄糖溶液(D5,pH<5)中测定生物效价时,肝素不稳定。也有人用生物效价和化学两种方法测定,发现肝素在D5中明显失活(40—55%),而在生理盐水中无影响。又有人报道,肝素在D5和生理盐水中6小时内效价均明显下降。但与上述结果相反的报道,认为在pH3.8—7.6、6.6—7.0、3.2—9.2,24小时内在D5中未出现效价下降。Coon最近指出,所以有这些差异均与测定方法、肝素的化学组份及其在混合介质中的降解作用有关。
The results of many years of research have been controversial for the stability of heparin as a function of pH. It is believed that heparin is unstable when measured for biological potency in a 5% glucose solution (D5, pH <5). Some people also use the biological potency and chemical determination of two methods and found that heparin in D5 was significantly inactivated (40-55%), but no effect in normal saline. It was also reported that heparin in D5 and saline within 6 hours were significantly lower titers. However, contrary to the above results, no decline in potency in D5 was observed at pH 3.8-7.6, 6.6-7.0, 3.2-9.2 for 24 hours. Coon recently pointed out that these differences are therefore related to the assay, the chemical composition of heparin and its degradation in mixed media.
