目的:探讨小柴胡汤(XCT)君臣药及其配伍提取物对感染CVB3m心肌细胞的抗病毒与保护作用,并试阐述机制。方法:制取3种提取物的含药血清,并测定体外对CVB3m毒力的影响。培养乳鼠心肌细胞建立心肌炎模型,从3个方面检测:应用MTT法检测含药血清对心肌细胞的保护率;检测各孔上清液中乳酸脱氢酶(LDH)及天冬氨酸氨基转移酶(AST)的释放量;RT-PCR法检测感染CVB3m的心肌细胞中CVB3m的含量。结果:黄芩提取物含药血清对CVB3m有直接灭活的作用。3个方面检测结果均显示:含药血清组与空白血清组比较,有显著性差异(P<0.01),黄芩组和配伍组分别与柴胡组比较,均有显著性差异(P<0.01),配伍组与黄芩组比较,无显著性差异。结论:黄芩提取物有直接灭活病毒的作用,黄芩和配伍提取物具有明显的抑制病毒复制和对病毒感染后细胞的保护作用,等量配伍提取物没有表现出作用增强的优势。
Objective: To investigate the anti-virus and protective effect of XCM and its compatibility extract on CVB3m-infected cardiomyocytes and to elucidate the mechanism. Methods: The drug-containing serums of three kinds of extracts were prepared and the effect on the virulence of CVB3m in vitro was determined. Myocardial cells were cultured to establish the model of myocarditis. Three aspects were tested: the protective rate of drug-containing serum on cardiomyocytes was detected by MTT assay; the activities of lactate dehydrogenase (LDH) and aspartate aminotransferase AST). The content of CVB3m in CVB3m-infected cardiomyocytes was detected by RT-PCR. Results: Scutellaria extract containing serum CVB3m direct inactivation. There were significant differences among the three groups (P <0.01). There were significant differences between Scutellariae Radix and compatibility group and Bupleurum group (P <0.01) , Compatibility group and baicalin group, no significant difference. CONCLUSION: Scutellaria baicalensis extract has direct inactivation of virus. Scutellaria baicalensis and compatibility extract have significant inhibitory effects on virus replication and cell protection after virus infection. Equivalent compatibility extract does not show the advantage of enhanced effect.
