目的研究细胞骨架稳定剂Jasplakinolide(JAS)对人脐静脉内皮细胞(human umbilical vein endothelial cells,HU-VECs)增殖、黏附、迁移、体外成血管能力等生物学功能的影响。方法 JAS(100 nmol·L-1)干预不同时间后,荧光显微镜观察HUVECs细胞骨架的改变。采用CCK-8法、黏附能力测定实验、改良的Boyden小室及Matrigel法分别检测其增殖、黏附、迁移及体外血管形成等功能性指标。结果 JAS对HUVECs细胞骨架的作用具有时间依赖性:100 nmol·L-1JAS处理1 h后细胞骨架明显变粗,荧光强度增加。而作用24 h后,细胞骨架聚集为团块状。JAS(100 nmol·L-1)可明显增加HUVECs的黏附、迁移及体外成血管能力,但对HUVECs增殖无明显影响。结论 JAS可通过稳定细胞骨架影响HUVECs的生物学特性。
Objective To investigate the effects of cytoskeletal stabilizer Jasplakinolide (JAS) on the biological functions of human umbilical vein endothelial cells (HU-VECs) such as proliferation, adhesion, migration and in vitro angiogenesis. Methods The changes of the cytoskeleton of HUVECs were observed under fluorescence microscope with JAS (100 nmol·L-1) for different time. CCK-8 assay, adhesion assay, modified Boyden chamber and Matrigel assay were used to detect the proliferation, adhesion, migration and angiogenesis in vitro. Results The effect of JAS on the cytoskeleton of HUVECs was time-dependent: the cytoskeleton became thicker and the fluorescence intensity increased after treated with 100 nmol·L-1 JAS for 1 h. After 24 h, the cytoskeleton aggregated into lumps. JAS (100 nmol·L-1) significantly increased the adhesion, migration and in vitro angiogenesis of HUVECs, but had no significant effect on the proliferation of HUVECs. Conclusion JAS can affect the biological characteristics of HUVECs by stabilizing the cytoskeleton.
