目的 :比较人正常、增生和癌变前列腺组织中PSP94和PSP5 7mRNA的表达情况。方法 :提取事故死亡的正常成年人前列腺组织及手术得到的增生和前列腺癌组织总RNA ,进行RT PCR ,其产物分别以PSP94和PSP5 7共有的外显子及PSP94外显子Ⅲ作探针进行Southernblotting分析 ;RT PCR产物经克隆后进行序列测定。结果 :三种前列腺组织中均有PSP94和PSP5 7mRNA的表达 ;人正常前列腺组织中的PSP94及PSP5 7的基因序列与增生前列腺组织和癌变前列腺组织中的完全一致。结论 :PSP5 7不是增生和癌变前列腺组织中PSP94mRNA可变剪切特有的产物
Objective: To compare the expression of PSP94 and PSP5 7 mRNA in normal, hyperplastic and cancerous prostate tissue. Methods: The normal adult prostatic tissue and the total proliferative prostate cancer tissue RNA extracted from the accident were collected and subjected to RT-PCR. The products were respectively exons shared by PSP94 and PSP5 7 and exon Ⅲ of PSP94 Southern blotting analysis; RT PCR products were cloned and sequenced. Results: The expression of PSP94 and PSP5 7 mRNA in the three kinds of prostatic tissues were identical. The sequence of PSP94 and PSP5 7 in human normal prostatic tissues were completely consistent with that in hyperplastic prostatic tissues and cancerous prostatic tissues. Conclusion: PSP5 7 is not a specific product of PSP94 mRNA variable shear in hyperplastic and cancerous prostate tissues
