基质金属蛋白酶是一类生物活性依赖于锌离子、有降解细胞外基质能力的内肽酶家族,最初在蝌蚪尾巴中发现。近年研究发现,基质金属蛋白酶可增强白细胞入侵能力、调控炎症过程及强度,可能存在双重双向的调节机制。现对近年基质金属蛋白酶在肺相关性疾病中若干机制和治疗方面研究的进展和前景做一综述,着重介绍基质金属蛋白酶在急性肺损伤、急性呼吸窘迫综合征、肺纤维化、肺移植等疾病中治疗和机制研究进展。最后讨论以基质金属蛋白酶作为治疗靶标存在的问题和临床应用展望。
Matrix metalloproteinases (MMPs) are a family of endopeptidases that are biologically active and dependent on zinc ions and have the ability to degrade extracellular matrix initially found in tadpole tails. Recent studies have found that matrix metalloproteinases can enhance the ability of leukocytes to invade, control the process and intensity of inflammation, and there may be a double bidirectional regulation mechanism. In recent years, the progress and prospects of several mechanisms and therapeutic aspects of matrix metalloproteinases in lung-related diseases are reviewed. The research progress of matrix metalloproteinases in acute lung injury, acute respiratory distress syndrome, pulmonary fibrosis and lung transplantation In the treatment and mechanism of research progress. Finally, we discuss the problems and clinical application of MMPs as therapeutic target.
