目的 :探讨SC - 5 812 5对HepG - 2细胞增殖和凋亡的作用及其分子机理。方法 :应用细胞培养、MTT、TUNEL、流式细胞光度术、琼脂糖凝胶电泳及Westernblot等方法研究SC - 5 812 5对HepG - 2细胞增殖和凋亡的作用及其分子机理。结果 :SC5 812 5抑制HepG - 2细胞的增殖、诱导其凋亡及引起G0 /G1期阻滞 ,S期抑制。并使P33cdk2 、P34 cdc2 、cyclinB1、cyclinE、Mpm - 2、Rb、PCNA 7种蛋白水平下降。结论 :SC5 812 5抑制HepG - 2细胞的增殖及诱导其凋亡 ,可能与P33cdk2 、P34 cdc2 、cyclinB1、cyclinE、Mpm - 2、Rb、PCNA 7种蛋白水平的下降有关。
Objective: To investigate the effect of SC - 5 812 5 on proliferation and apoptosis of HepG - 2 cells and its molecular mechanism. Methods: The effects of SC - 5 812 5 on proliferation and apoptosis of HepG - 2 cells were studied by cell culture, MTT, TUNEL, flow cytometry, agarose gel electrophoresis and Western blotting. Results: SC5 812 5 inhibited the proliferation of HepG - 2 cells, induced apoptosis and induced G0 / G1 phase arrest and S phase inhibition. P33cdk2, P34 cdc2, cyclinB1, cyclinE, Mpm - 2, Rb and PCNA decreased. Conclusion: The inhibitory effect of SC5 812 5 on the proliferation and apoptosis of HepG - 2 cells may be related to the decrease of 7 protein levels of P33cdk2, P34 cdc2, cyclinB1, cyclinE, Mpm - 2, Rb and PCNA.
