目的:研究天宁饮对硝酸甘油所致偏头痛大鼠行为症状及三叉神经核尾部c-fos蛋白表达的影响,从动物实验角度证实天宁饮的疗效及其作用机理。方法:将60只雌雄各半SD大鼠随机分为空白对照组、偏头痛模型组、阳性对照组(盐酸氟桂利嗪组)、天宁饮高、中、低剂量组,每组10只。除空白对照组外,其他组采用皮下注射硝酸甘油(10 mg/kg)复制偏头痛大鼠模型,造模后30 min分别给予蒸馏水及药物灌胃,观察各组大鼠耳红开始、结束时间及不同时间挠头、爬笼次数;同时观察模型大鼠三叉神经核尾部c-fos蛋白表达情况。结果:与空白组比较,模型组出现耳红现象,挠头、爬笼次数显著性增加(P<0.01),三叉神经核尾部c-fos蛋白表达亦显著性增加(P<0.01);与模型组比较,阳性对照组(盐酸氟桂利嗪组)、天宁饮高、中、低剂量组均可明显缩短模型大鼠耳红时间,抑制模型大鼠挠头、爬笼次数(P<0.05),显著性抑制模型大鼠三叉神经核尾部c-fos蛋白表达(P<0.01)。结论:天宁饮可有效治疗硝酸甘油型所致大鼠偏头痛,其作用机制与抑制模型大鼠三叉神经核尾部c-fos蛋白表达有关。

Results: Compared with the blank group, the model group showed ear red phenomenon, the number of scratching head and climbing cage increased significantly (P <0.01), while the expression of c-fos protein increased significantly in the tail of the trigeminal nucleus (P <0.01) Compared with the control group (Flunarizine Hydrochloride Group) and Tianning Decoction high, medium and low dose groups, the ear time of model rats could be significantly shortened and the numbers of scratching head and climbing cage of model rats were inhibited (P <0.05) Significantly inhibited c-fos protein expression in the tail of the trigeminal nucleus in model rats (P <0.01). CONCLUSION: Tianning Decoction can effectively treat migraine induced by nitroglycerin in rats and its mechanism is related to inhibiting the expression of c-fos protein in the tail of the trigeminal nucleus in model rats.
