背景:间充质干细胞分泌的因子所形成的的微环境能够抑制肿瘤细胞的恶性增殖,而盐霉素也对肿瘤细胞生长有抑制作用。目的:探讨盐霉素联合脐血间充质干细胞干预对乳腺癌MCF-7细胞的影响及作用机制,为乳腺癌的治疗寻找新的靶点和治疗策略。方法:选取对数生长的MCF-7乳腺癌细胞,将其随机分为对照组、脐血间充质干细胞组及联合组。对照组细胞不做任何处理,干预组加入脐血间充质干细胞悬液,联合组细胞同时加入脐血间充质干细胞悬液和盐霉素。结果与结论:干预48 h后,与对照组及脐血间充质干细胞组相比,联合组乳腺癌MCF-7细胞的增殖与侵袭能力抑制作用更为明显;细胞中POSTN蛋白的表达水平也较低。提示脐血间充质干细胞与盐霉素联合干预能有效抑制乳腺癌MCF-7细胞的增殖与迁移,为乳腺癌的治疗提出一种新的治疗方法。
BACKGROUND: The microenvironment formed by the factors secreted by mesenchymal stem cells can inhibit the malignant proliferation of tumor cells, while salinomycin also inhibits tumor cell growth. OBJECTIVE: To investigate the effect of salinomycin combined with umbilical cord blood mesenchymal stem cells on breast cancer cell line MCF-7 and its mechanism of action, so as to find a new target and treatment strategy for the treatment of breast cancer. Methods: Logarithmic growth of MCF-7 breast cancer cells were selected and randomly divided into control group, umbilical cord blood mesenchymal stem cells group and combination group. Control group cells without any treatment, the intervention group added to the umbilical cord blood mesenchymal stem cell suspension, the combined group of cells while adding cord blood MSCs suspension and salinomycin. RESULTS AND CONCLUSION: Compared with the control group and umbilical cord blood mesenchymal stem cells group, the inhibitory effect on proliferation and invasion ability of breast cancer MCF-7 cells in the combined group was more obvious after 48 h intervention.
