目的黄连素(BBR)对糖尿病及血脂异常的作用仍未完全阐明。该研究旨在研究BBR对高脂饮食诱导的肥胖(DIO)大鼠模型的作用机制。探索其与内质网(ER)应激及炎症反应的作用。方法高脂饮食喂养的大鼠给予不同浓度的BBR及10 mg/(kg.d)的匹格列酮。监测血糖、胰岛素及生化指标。使用实时PCR分析内质网应激的分子生物学标志物及炎症相关基因。结果通过空腹血糖和血脂水平可见BBR降血糖及血脂的效应。高剂量BBR组,炎症基因TNF-α、IL-6及ER应激相关基因CHOP和eIF下降。结论 BBR对DIO大鼠代谢改善作用可能与ER应激及炎症反应的降低有关。
The purpose of the role of berberine (BBR) on diabetes and dyslipidemia has not been fully elucidated. The aim of this study was to investigate the mechanism of action of BBR on a model of obesity (DIO) induced by a high-fat diet. To explore its role in endoplasmic reticulum (ER) stress and inflammatory response. Methods Rats fed high-fat diet were given different concentrations of BBR and 10 mg / (kg.d) pioglitazone. Monitoring blood glucose, insulin and biochemical indicators. Real-time PCR analysis of endoplasmic reticulum stress molecular biomarkers and inflammation related genes. Results The effects of BBR on lowering blood glucose and blood lipids were observed by fasting blood glucose and blood lipid levels. High-dose BBR group, inflammatory genes TNF-α, IL-6 and ER stress-related genes CHOP and eIF decreased. Conclusion BBR may improve the metabolism of DIO rats and may be related to the decrease of ER stress and inflammatory reaction.
