从T细胞亚群变化的角度探讨补肾活血汤治疗免疫性卵巢早衰的免疫调控机制。对B6AF1小鼠进行皮下注射ZP3多肽,建立自身免疫性卵巢早衰小鼠模型,用补肾活血汤进行灌胃治疗,实验结束后,计算脏器指数,对卵巢进行H&E染色和ZP3免疫荧光染色来检测卵巢组织形态变化和透明带抗体分布,流式细胞术检测脾脏中CD4~+T细胞、CD8~+T细胞、CD4~+CD25~+Treg细胞和CD4~+IL-17~+Th17细胞比例。结果显示补肾活血汤能够提高免疫性卵巢早衰小鼠的卵巢指数,减少卵巢组织中炎症细胞浸润,增加卵泡的数量,减少卵巢组织中透明带抗体的产生;补肾活血汤能够下调脾脏中CD8~+T细胞和Th17细胞的比例,上调脾脏中CD4~+T细胞和Treg细胞的比例。表明补肾活血汤能够通过调节T细胞亚群的平衡,减轻炎症反应的程度,从而改善免疫性卵巢早衰。
Study on the Immune Regulation Mechanism of Bushen Huoxue Decoction on Premature Ovarian Failure from the Perspective of T Cell Subsets. B6AF1 mice were injected subcutaneously with ZP3 peptide to establish autoimmune premature ovarian failure model in mice. The mice were treated with Bushen Huoxue Decoction. After the experiment, the index of organ was calculated, H & E staining and ZP3 immunofluorescence staining were used to detect the ovary The morphological changes of ovarian tissue and antibody distribution of zona pellucida were observed. The proportion of CD4 ~ + T cells, CD8 ~ + T cells, CD4 ~ + CD25 ~ + Treg cells and CD4 ~ + IL-17 ~ + Th17 cells in spleen were detected by flow cytometry. The results showed that Bushen Huoxue Decoction can improve the ovarian index of premature ovarian failure mice, reduce the infiltration of inflammatory cells in ovarian tissue, increase the number of follicles and reduce the production of zona pellucida in ovary tissue. Bushen Huoxue Decoction can down-regulate the expression of CD8 + The proportion of T cells and Th17 cells increased the proportion of CD4 ~ + T cells and Treg cells in the spleen. That Bushen Huoxue Decoction can regulate the balance of T cell subsets, reduce the degree of inflammatory response, thereby improving immune premature ovarian failure.
