杜鹃是杜鹃花科(Ericaeae)杜鹃花属(Rhododendron)植物,具有较高的观赏价值和生态维持价值。该研究采用EST-SSR标记对湖北龟峰山32个天然杜鹃花样品进行遗传多样性分析,旨在为龟峰山杜鹃的育种研究及新品种选育提供理论基础。7对EST-SSR引物共扩增出31条多态性条带,平均每个位点的等位基因数为4.43个,平均观察杂合度Ho和期望杂合度He的值分别为0.679 58和0.723 14。该研究开发的EST-SSR标记不仅丰富了现有的SSR数据库,也为后续分子标记辅助育种、遗传多样性和遗传结构分析、亲缘关系分析奠定了基础。
Rhododendron is the genus Rhododendron of Ericaeae, which has high ornamental value and ecological preservation value. In this study, genetic diversity of 32 natural rhododendron samples from Guifeng Mountain in Hubei Province was analyzed by EST-SSR marker, which provided a theoretical basis for breeding of Cuckoo rhododendron and selection of new varieties. Seven pairs of EST-SSR primers amplified a total of 31 polymorphic bands with an average of 4.43 alleles per locus. The mean observed heterozygosity Ho and expected heterozygosity He were 0.679 58 and 0.723, respectively 14. The EST-SSR markers developed in this study not only enriched the existing SSR database, but also laid the foundation for subsequent molecular marker-assisted breeding, genetic diversity and genetic structure analysis and genetic relationship analysis.
