目的探讨microRNA-4731-3p(miR-4731-3p)在肺癌血浆中的表达及其临床意义。方法采用实时荧光定量PCR(qRT-PCR)方法检测40例肺癌患者和40例健康人(作为对照)血浆miR-4731-3p的表达水平,分析其与临床病理参数和预后的相关性。绘制受试者工作特征曲线(ROC),评价血浆miR-4731-3p表达水平对原发性肺癌的诊断价值。结果肺癌血浆中miR-4731-3p的表达水平(0.11±0.05)显著高于健康人(0.05±0.03)(P<0.001);miR-4731-3p表达水平与癌胚抗原(CEA)正相关(P<0.05),与患者性别、年龄、吸烟、病理类型、肿瘤大小、临床分期、远处转移均无明显相关性(P>0.05)。血浆miR-4731-3p在ROC曲线下面积(AUC)为0.835(95%CI=0.750~0.921)。当miR-4731-3p临界值取0.048时,对肺癌诊断的灵敏度为0.950,特异度为0.575。KaplanMeier分析显示肺癌血浆中miR-4731-3p低表达者生存时间明显高于高表达组(P<0.05)。结论血浆miR-4731-3p在肺癌血浆中表达上调,可作为肺癌诊断及预后判断的潜在分子标志物。`
Objective To investigate the expression of microRNA-4731-3p (miR-4731-3p) in plasma of lung cancer and its clinical significance. Methods Real-time quantitative PCR (qRT-PCR) was used to detect the plasma levels of miR-4731-3p in 40 lung cancer patients and 40 healthy controls (as controls), and its correlation with clinicopathological parameters and prognosis was analyzed. The receiver operating characteristic curve (ROC) was drawn to evaluate the diagnostic value of plasma miR-4731-3p expression in primary lung cancer. Results The expression level of miR-4731-3p in lung cancer was significantly higher than that in healthy controls (0.11 ± 0.05 vs 0.05 ± 0.03, P <0.001). The expression of miR-4731-3p was positively correlated with carcinoembryonic antigen (CEA) P <0.05). There was no significant correlation with gender, age, smoking, pathological type, tumor size, clinical stage and distant metastasis (P> 0.05). The area under the ROC curve (AUC) of plasma miR-4731-3p was 0.835 (95% CI = 0.750-0.921). When the critical value of miR-4731-3p is taken as 0.048, the diagnostic sensitivity for lung cancer is 0.950 and the specificity is 0.575. KaplanMeier analysis showed that the survival time of lung cancer plasma miR-4731-3p low expression was significantly higher than the high expression group (P <0.05). Conclusion Plasma miR-4731-3p is upregulated in plasma of lung cancer and may be used as a potential molecular marker for the diagnosis and prognosis of lung cancer. `
