本文报道了2种分别由人巨细胞病毒(hCMV)启动子和SV40早期启动子控制的人Ⅸ因子cDNA质粒pCMV─Ⅸ,pKG5─Ⅸ直接注射小鼠骨骼肌内,在肌细胞中可产生Ⅸ因子蛋白,并分泌至血液中,在注射后约第10d表达量最高,最高含量可达25ng/ml。为采用直接注射法进行遗传病基因治疗提供1种可能。
In this paper, two kinds of human factor IX cDNA plasmids pCMV-IX and pKG5-IX which are respectively controlled by human cytomegalovirus (hCMV) promoter and SV40 early promoter are reported to be directly injected into the skeletal muscle of mice and IX Factor protein and secreted into the blood, the highest expression level of about 10d after injection, the maximum content of up to 25ng / ml. It is possible to use gene therapy for direct gene therapy.
