目的:采用反转录聚合酶链反应(RT-PCR)的检测方法观察细胞因子肿瘤坏死因子(TNF-αmRNA)的含量,探讨膝骨关节炎的发病机制,研究单味中药骨碎补通过治疗膝骨关节炎的作用机理,并加以讨论、分析。方法:将24只成年新西兰大耳白兔,随机分为4组,空白组6只,模型组6只,治疗组(骨碎补组)6只,对照组(维固力组)6只。兔左后肢伸直位管型石膏固定6周后得到兔膝骨关节炎的模型。通过灌胃给药单味中药骨碎的补水煎浓缩液以及西药维固力的浓缩液4周。采用耳缘静脉空气栓塞方法处死各组动物,通过外科手术取下其患肢股骨髁(大小约1 cm×1 cm),并分离关节软骨,保留其滑膜,用Trizol液浸泡,4℃冷藏保存,用于RTPCR的检测。本实验结果采用SPSS 13.0软件包进行相关分析,P<0.05有统计学意义。结果:单味中药骨碎补能够影响兔膝骨关节炎模型关节滑膜中的细胞因子的表达,给药组滑膜中TNF-α的表达明显低于模型组,差异有统计学意义(P<0.05)。结论:单味中药骨碎补可通过降低关节滑膜中TNF-α的表达,从而保护关节软骨免受损伤以及破坏,说明骨碎补对膝骨关节炎有比较显著的防治效果。
Objective: To detect the content of tumor necrosis factor (TNF-αmRNA) by reverse transcription-polymerase chain reaction (RT-PCR) and explore the pathogenesis of knee osteoarthritis. The mechanism of knee osteoarthritis, and to discuss and analyze. Methods: Twenty-four adult New Zealand white rabbits were randomly divided into 4 groups, 6 in the blank group, 6 in the model group, 6 in the treatment group (Osamu group) and 6 in the control group (Vitiligo group). Rabbit knee osteoarthritis model was obtained after 6 weeks of immobilization of left hind limb extension type plaster. By gavage single Chinese herbal medicine bone loss of water and decoction concentrate and Western Wei force solid concentrate for 4 weeks. Animals in each group were sacrificed by ear vein occlusion. The femoral condyles (about 1 cm × 1 cm in size) were removed surgically and the articular cartilage was removed. The synovium was preserved and soaked in Trizol solution for 4 ℃ Save for RTPCR detection. The experimental results using SPSS 13.0 package for correlation analysis, P <0.05 was statistically significant. Results: The single herb of traditional Chinese medicine, Osteoarthritis, could influence the expression of cytokines in synovium of rabbit knee osteoarthritis model. The expression of TNF-α in the synovial membrane of the treated group was significantly lower than that of the model group (P <0.05). Conclusion: The traditional Chinese medicine Drynaria can protect the articular cartilage from injury and damage by decreasing the expression of TNF-α in synovium, indicating that Osteopractic has significant preventive and therapeutic effects on knee osteoarthritis.
