目的:观察黄连总生物碱(Coptis alkaloids,CA)对糖尿病肾病(Diabetic Nephropathy,DN)大鼠的保护作用。方法:大鼠适应性饲养1周,禁食不禁水12h后,按60mg/kg剂量一次性腹腔注射链脲佐菌素(streptozotocin,STZ)的方法复制大鼠糖尿病模型。72h后尾静脉取血,测血糖,选取血糖值≥16.7mmol/L大鼠100只,随机分为模型组、黄连总生物碱低(50mg/kg)、中(100mg/kg)、高剂量(200mg/kg)组,阳性药格列齐特(20mg/kg)组,另取未造模大鼠10只为正常组。给药组分别灌胃给予黄连总生物碱、格列齐特,正常组和模型组给予等容量溶媒,每天1次,连续8周。末次给药后,禁食12h处置动物,取相应指标检测:检测大鼠空腹血糖(fasting blood glucose,FBG)水平;HE常规染色,光镜下观察大鼠肾脏病理学变化情况;全自动生化仪检测血肌酐(serum creati-nine,Scr)、血尿素氮(boold urea nitrogen,BUN)含量;放射免疫分析法检测胰岛素(Insulin,Ins)、C肽(C-PePtide,C-P)、肿瘤坏死因子(Tumor necrosis factor,TNF-α)的含量。结果:实验结果显示,与模型组相比,黄连总生物碱(50mg/kg、100mg/kg、200mg/kg)组能降低大鼠FBG、TNF-α、BUN、Scr水平,升高Ins;黄连总生物碱(100mg/kg、200mg/kg)组能升高C-P含量;病理组织学检查亦提示,黄连总生物碱能够改善肾脏病理组织学损伤程度。结论:黄连总生物碱对糖尿病肾病大鼠具有一定的保护作用。
Objective: To observe the protective effect of Coptis alkaloids (CA) on diabetic nephropathy (DN) rats. Methods: The rats were reared for 1 week. After fasting for 12 hours, rats were dosed with 60 mg / kg streptozotocin (STZ) intraperitoneally to induce the diabetic model. Blood samples were taken from the tail vein 72 h after the test to determine the blood glucose level. 100 rats with a blood glucose level ≥16.7 mmol / L were randomly divided into model group (50 mg / kg), medium (100 mg / kg) 200mg / kg) group, the positive drug Gliclazide (20mg / kg) group, another non-model rats 10 normal group. Administration group were given gavage of total alkaloids, gliclazide, normal group and model group were given the same volume of vehicle, once a day for 8 weeks. After the last administration, the rats were fasted for 12 hours, and the corresponding indexes were detected: the level of fasting blood glucose (FBG) in rats was detected; the pathological changes of kidney in rats were observed by HE staining and light microscope; Serum creati-nine (Scr) and blood urea nitrogen (BUN) were detected by radioimmunoassay. Insulin (Ins), C-Peptide (CP) and tumor necrosis factor Tumor necrosis factor, TNF-α). Results: The results showed that the total alkaloids (50mg / kg, 100mg / kg, 200mg / kg) of Coptis chinensis could decrease the level of FBG, TNF-α, BUN and Scr, Total alkaloids (100mg / kg, 200mg / kg) group can increase the CP content; histopathological examination also suggests that Coptis alkaloids can improve renal histopathological damage. Conclusion: Coptis alkaloids have some protective effects on diabetic nephropathy rats.
