目的探讨柚皮素对人肺成纤维细胞生成血管内皮生长因子(VEGF)的影响,并研究其可能的机制。方法培养人胚肺成纤维细胞,分为对照组、香烟提取物(CSE)组和柚皮素干预组。柚皮素干预组分别与低、中、高剂量的柚皮素共孵育2 h后,与CSE组一起加入CSE,调整终浓度柚皮素5μmol/L、10μmol/L和20μmol/L,CSE终浓度5%。培养24、48及72 h后,ELISA法测定各组VEGF含量,采用Western blot法观察各组Smad3及磷酸化Smad3(p-Smad3)的表达情况。结果在人胚肺成纤维细胞中,柚皮素能抑制CSE诱导的血管内皮生长因子生成。CSE能显著提高p-Smad3水平,而添加了柚皮素后p-Smad3明显减少,并随柚皮素剂量的增加而p-Smad3水平愈低。结论柚皮素能够抑制Smad3的磷酸化,从而抑制CSE诱导的血管内皮生长因子的分泌。
Objective To investigate the effect of naringenin on human lung fibroblast-derived vascular endothelial growth factor (VEGF) and its possible mechanism. Methods Human embryonic lung fibroblasts were cultured and divided into control group, cigarette smoke extract (CSE) group and naringenin intervention group. Naringenin intervention groups were incubated with low, medium and high dose of naringenin for 2 h, then added CSE together with CSE group to adjust the final concentration of naringenin 5μmol / L, 10μmol / L and 20μmol / L, CSE terminal Concentration of 5%. After culturing for 24, 48 and 72 hours, the content of VEGF in each group was determined by ELISA. The expression of Smad3 and p-Smad3 in each group was observed by Western blot. Results In human embryonic lung fibroblasts, naringenin can inhibit CSE-induced vascular endothelial growth factor. CSE could significantly increase the level of p-Smad3, while p-Smad3 decreased significantly after addition of naringenin, and the level of p-Smad3 was lower with the increase of dosage of naringenin. Conclusion Naringenin can inhibit the phosphorylation of Smad3, thereby inhibiting CSE-induced vascular endothelial growth factor secretion.
