目的 研究纤维连接蛋白 (fibronectin ,FN)及其受体整合素α5β1在肺纤维化中的作用。方法 应用Northern印迹杂交和免疫细胞化学技术。结果 经FN作用 2h ,肺成纤维细胞α5β1和FNmRNA的杂交信号均增强 ,6h达最高值 ;FN作用 6、1 2、2 4h ,免疫细胞化学染色显示整合素α5β1、FN蛋白的表达增强 ;经α5单克隆抗体预作用 ,FN作用组的肺成纤维细胞整合素α5β1和FN蛋白表达减弱 ,其抑制作用依次分别在 6h(α5β1)、1 2h(FN)最明显。结论 FN能直接刺激肺成纤维细胞合成FN或通过上调整合素α5β1的表达来促进自身合成FN。
Objective To investigate the role of fibronectin (FN) and its receptor integrin α5β1 in pulmonary fibrosis. Methods Northern blotting and immunocytochemistry were used. Results After FN treatment for 2h, the hybridization signals between α5β1 and FN mRNA in lung fibroblasts were all increased, reaching the highest value at 6h. The FN effect was 6,1,224 h, and the expression of integrin α5β1 and FN protein was enhanced by immunocytochemical staining. α5 monoclonal antibody preconditioning. The expression of integrin α5β1 and FN in lung fibroblasts decreased with FN treatment. The inhibitory effect was the most obvious at 6h (α5β1) and 12h (FN) respectively. Conclusion FN can directly stimulate fibroblasts to synthesize FN or promote the synthesis of FN by up-regulating the expression of integrin α5β1.
