目的探讨5-羟色胺(5-HT)信号传导通路相关基因位点多态性与驾驶员事故倾向性的关联。方法研究对象来自2014年在广西工人医院体检的驾驶员,通过问卷调查将过去5年中发生≥3次责任交通事故的驾驶员纳入病例组,对照组按年龄、性别、驾驶车型、驾龄与病例组进行成组匹配;对G蛋白β3亚基(GNβ3)基因rs5443位点、脑源性神经营养因子(BDNF)基因rs6265位点和反应元件结合蛋白(CREB)基因rs6740584位点进行单核苷酸多态性(SNP)基因分型。结果 rs5443位点基因型频率分布在病例组与对照组分布有统计学差异(χ2=7.20,P=0.027),rs6265和rs6740584位点基因型频率在2组分布均无统计学差异(均P>0.05);携带rs5443位点突变基因型TT可降低事故倾向性风险,rs6265和rs6740584位点多态性在调整前后与驾驶员事故倾向性均无统计学关联。结论 GNβ3基因rs5443多态性与驾驶员的事故倾向性可能有关联。
Objective To investigate the association between 5-HT signaling pathway-related gene polymorphisms and driver accident predisposition. Methods The subjects were from the medical workers in Guangxi Workers’ Hospital in 2014. The surveyed drivers included ≥3 drivers who were responsible for traffic accidents in the past 5 years. The control subjects were divided into groups according to age, gender, driving style, driving experience and cases Groups were grouped and matched. Single nucleotide analysis was performed on rs5443 of GNβ3 gene, rs6265 of BDNF gene and rs6740584 of CREB gene. Polymorphism (SNP) genotyping. Results There was a statistically significant difference in genotype distribution between rs5443 locus and control group (χ2 = 7.20, P = 0.027). There was no significant difference in rs6265 and rs6740584 genotype distribution between the two groups (all P> 0.05). Incidence of genotype TT at rs5443 locus reduced the risk of accidental predisposition. There was no significant correlation between rs6265 and rs6740584 locus polymorphism before and after driver adjustment. Conclusion The rs5443 polymorphism of GNβ3 gene may be related to driver’s accident tendency.
