目的观察水蛭素对喉癌荷瘤小鼠的抑瘤作用。方法将40只小鼠随机分为4组,每组10只,小鼠左前腋窝下皮下种瘤建立实体瘤模型,阳性对照组以氟尿嘧啶20mg/kg腹腔注射;水蛭素高剂量组以2.5g/(kg·d)灌胃;阴性对照组以生理盐水0.1m L/(10g·d)灌胃;水蛭素低剂量组以1.2g/(kg·d)灌胃。连续用药14 d后处死小鼠取荷瘤鼠肿块称重,并将瘤组织包埋、固定、切片、染色等,计算抑瘤率,利用Image-Pro Plus 6.0图像分析软件检测瘤组织中C-met的表达。结果水蛭素高、低剂量组对小鼠喉癌细胞均有明显的抑制作用,抑瘤率分别为40.1%、26.92%,与对照组比较差异有统计学意义(P<0.05);4组中C-met均呈阳性表达,水蛭素高、低剂量组C-met的表达均下调,与阴性对照组比较差异有统计学意义(P<0.05)。结论水蛭素对小鼠喉癌细胞抑制作用显著。
Objective To observe the inhibitory effect of hirudin on tumor-bearing mice with laryngocarcinoma. Methods Forty mice were randomly divided into 4 groups with 10 animals in each group. Solid tumor models were established in subcutaneous anterior subcapsular anomaly in mice. The positive control group was injected intraperitoneally with 20 mg / kg fluorouracil. The high-dose Hirudin group was given 2.5 g / (kg · d). The rats in the negative control group were given gavage with saline 0.1m L / (10g · d), and the hirudin low dose group was gavaged with 1.2g / (kg · d). The mice were sacrificed after 14 days of continuous administration. The tumor-bearing mice were weighed, and the tumor tissues were embedded, fixed, sectioned and stained to calculate the tumor inhibition rate. Image-Pro Plus 6.0 image analysis software was used to detect C- met expression. Results The hirudin high and low dose groups had significant inhibitory effects on the laryngeal carcinoma cells in mice with the inhibitory rates of 40.1% and 26.92%, respectively, which were significantly different from those of the control group (P <0.05) C-met expression were positive, hirudin high and low doses of C-met expression were down regulation, compared with the negative control group, the difference was statistically significant (P <0.05). Conclusion hirudin has a significant inhibitory effect on mouse laryngeal carcinoma cells.
