[目的]探讨肝癌(HCC)组织中组织蛋白酶B(CB)和CB mRNA的表达和作用。[方法]应用免疫组织化学和RT-PCR方法测定71例肝组织标本CB及CB mRNA,其中对照(C)组12例,HCC组34例和HCC癌旁组织(PTT)组25例,对比分析CB、CB mRNA与肝癌临床病理因素的关系。[结果]HCC组CB的平均吸光度值和CBmRNA条带的光密度比值均明显高于C组和PTT组(均P<0.01),PTT组与C组间均无显著差异(P>0.05)。CB在包膜不完整,有肝内转移和肝外转移HCC中的表达分别明显高于包膜完整,无肝内转移和肝外转移者(P<0.05或<0.01);与肿瘤的大小无关(P>0.05);随组织分化降低,均有增高趋势,但无统计学意义。[结论]HCC组织CB及其基因均呈现高表达,可能在HCC的发生、发展,尤其是侵袭和转移过程中发挥重要的作用。
[Objective] To investigate the expression and effect of cathepsin B (CB) and CB mRNA in hepatocellular carcinoma (HCC). [Method] Immunohistochemistry and RT-PCR were used to determine CB and CB mRNA in 71 cases of liver tissue. Among them, 12 cases in control group (C), 34 cases in HCC group and 25 cases in HCC adjacent tissue group (PTT) Relationship between CB, CB mRNA and clinicopathological factors of hepatocellular carcinoma. [Results] The mean absorbance values of CB and CB mRNA bands in HCC group were significantly higher than those in C group and PTT group (all P <0.01). There was no significant difference between PTT group and C group (P> 0.05). The expression of CB in HCC with incomplete envelopment, intrahepatic metastasis and extrahepatic metastasis was significantly higher than that with complete envelopment, without intrahepatic metastasis and extrahepatic metastasis (P <0.05 or <0.01), but not with tumor size (P> 0.05). With the decrease of tissue differentiation, there was an increasing tendency, but no statistical significance. [Conclusion] The high expression of CB and its genes in HCC tissues may play an important role in the occurrence and development of HCC, especially in invasion and metastasis.
