目的观察鞘内注射米诺环素对甲醛致痛大鼠的行为学和脊髓背角c-fos表达的影响。方法 24只雄性SD大鼠随机分为正常对照组、生理盐水组和米诺环素组,每组8只。对照组不做任何干预,生理盐水组和米诺环素组在右足底注射5%甲醛前30 min分别鞘内注射生理盐水20μl、米诺环素50μg。记录足底注射甲醛后1 h内每5 min的舔爪和缩腿时间,然后免疫组化观察脊髓背角c-fos的表达。结果米诺环素鞘内注射明显减少甲醛诱导的缩腿和舔爪时间,显著减少注射侧脊髓背角c-fos的表达(P<0.05或<0.01)。结论米诺环素鞘内注射能减少甲醛诱导的疼痛反应和脊髓背角c-fos的表达。
Objective To observe the effects of intrathecal minocycline on the behavior and the expression of c-fos in spinal dorsal horn induced by formaldehyde in rats. Methods Twenty-four male SD rats were randomly divided into normal control group, normal saline group and minocycline group, with 8 rats in each group. The control group did not make any intervention. In the saline group and the minocycline group, intrathecal injection of 20μl normal saline and 50μg minocycline respectively 30 min before the injection into the right plantar 5% formaldehyde. Recording lumbar paw withdrawal and leg reduction every 5 min within 1 h after injection of formaldehyde in the plantar, and then immunohistochemical observation of c-fos expression in spinal dorsal horn. Results Minocycline intrathecal injection significantly reduced the time of formalin-induced leg reduction and licking, and significantly decreased the expression of c-fos in the dorsal horn on the injected side (P <0.05 or <0.01). Conclusion Minocycline intrathecal injection can reduce formaldehyde-induced pain response and c-fos expression in spinal dorsal horn.
