目的 观察微量元素硒 (Se)对大鼠肝细胞、储脂 (Ito)细胞过氧化脂质和细胞外基质 (ECM)产生的影响。方法 采用原位灌注方法分离大鼠肝细胞和 Ito细胞。结果 培养肝细胞、Ito细胞有一定水平的基础丙二醛(MDA)产生和 ECM分泌。Se可提高肝细胞、Ito细胞谷胱甘肽过氧化物酶 (GSH- Px)活性 ,抑制基础 MDA产生 ,减少 3H-脯氨酸 (3H- Pro)掺入、 型前胶原 (PC )分泌及 Ito细胞透明质酸 (HA)分泌。结论 Se可抑制肝细胞和 Ito细胞过氧化脂质产生和 ECM分泌
Objective To observe the effect of trace element selenium (Se) on the production of lipid peroxidation and extracellular matrix (ECM) in hepatocytes and Ito cells in rats. Methods The rat hepatocytes and Ito cells were isolated by in situ perfusion. Results In cultured hepatocytes, Ito cells had some level of basal MDA and ECM secretion. Se can increase the activity of GSH-Px in hepatocytes and Ito cells, inhibit the basal MDA production and decrease the incorporation of 3H-Pro, the secretion of procollagen (PC) and Ito cells hyaluronic acid (HA) secretion. Conclusion Se can inhibit lipid peroxidation and ECM secretion in hepatocytes and Ito cells
