目的:优选水牛角中总氨基酸水解提取的最佳工艺。方法:以提取物中总氨基酸的含量及提取物对凝血酶引起纤维蛋白凝固的时间为考察指标,采用正交试验,以酸水浓度、酸水用量及水解时间为因素,优选最佳水解条件。结果:水牛角加8倍量4 mol.L-1硫酸回流加热水解提取6 h为水牛角最佳水解工艺。结论:优选的水解工艺提取的氨基酸含量较高,工艺稳定可行。
Objective: To optimize the optimum extraction of total amino acid in buffalo horn. Methods: Taking the content of total amino acids in extract and the time of fibrin coagulation induced by thrombin as indexes, orthogonal test was used to determine the optimum hydrolysis conditions by using acid water concentration, acid water dosage and hydrolysis time as factors . Results: Buffalo horn plus 8 times the amount of 4 mol.L-1 sulfuric acid heating hydrolysis extraction 6 h buffalo horn hydrolysis process. Conclusion: The preferred hydrolysis process has a high content of amino acids and the process is stable and feasible.
