目的:初步探讨降压益肾颗粒对自发性高血压大鼠(SHR)的降压效果及对血浆ET-1和TGF-β1含量的影响。方法:选择8周龄雄性SHR50只,随机分为SHR模型组、降压益肾颗粒低、中、高剂量组、盐酸贝那普利组,同时以同源性Wistar大鼠10只作为正常对照组,药物干预12周。每周用无创尾动脉套袖法测量各组大鼠血压;第8周和12周后取血,测定血清中ET-1和TGF-β1含量。结果:SHR模型组第8周和12周血浆中ET-1和TGF-β1含量明显升高,与Wistar组比较具有显著性差异(P<0.01),降压益肾颗粒可明显降低SHR血压和血清中ET-1和TGF-β1含量,与SHR模型组比较具有显著性差异(P<0.01),结论:降压益肾颗粒对SHR有较好降压作用,其降压机制可能与降低SHR血浆ET-1和TGF-β1含量有关。
Objective: To investigate the antihypertensive effect of Jiangya Yishen Granule on spontaneously hypertensive rats (SHR) and its effect on plasma ET-1 and TGF-β1 levels. Methods: A total of 8 SHR50 male SHR rats were randomly divided into sham-operated SHR model group, low-dose, middle-dose and high-dose shanghai yishen granules group and benazepril hydrochloride group. Meanwhile, 10 Wistar rats were used as normal control Group, drug intervention for 12 weeks. The blood pressure of each group was measured by noninvasive tail artery sleeve method every week. Blood was taken at 8th week and 12th week, and the levels of ET-1 and TGF-β1 in serum were measured. Results: Compared with Wistar group, the levels of ET-1 and TGF-β1 in SHR model group were significantly increased at 8th week and 12th week (P <0.01). The blood pressure of SHR model group was significantly lower than that of Wistar group The levels of ET-1 and TGF-β1 in serum of SHR group were significantly lower than those of SHR group (P <0.01). Conclusion: JJH can depress SHR effectively, Plasma ET-1 and TGF-β1 levels.
