目的 探讨RASSF1A(RAS相关区域家族 1A)基因转录表达与肺癌发生、发展的关系。方法 采用逆转录 聚合酶链反应 (RT PCR)的方法检测 4 7例肺癌患者癌组织及相应癌周正常组织中RASSF1AmRNA的表达水平。结果 (1)RASSF1AmRNA在所有正常组织中表达 ,而在肺癌癌组织中存在较高的表达缺失率 (5 3 2 % )。 (2 )淋巴结转移阳性者癌组织中RASSF1AmRNA表达缺失率 (70 4 % )明显高于淋巴结转移阴性者 (30 0 % ) (P <0 0 1) ;(3)RASSF1AmRNA表达缺失率与肺癌分期有关 ,越晚病例RASSF1A表达缺失率越高 (P <0 0 5 ) ;(4 )RASSF1AmRNA表达缺失与肿瘤组织学类型、肿瘤分化程度及吸烟指数未见显著相关 (P >0 0 5 )。结论 RASSF1A转录表达缺失 ,它可能参与调控肺癌的发生、发展过程 ,并进而影响其生物学特点及预后。
Objective To investigate the relationship between RASSF1A gene transcription and gene expression and the occurrence and development of lung cancer. Methods RT-PCR was used to detect the expression of RASSF1A mRNA in 47 cases of lung cancer and corresponding normal tissues. Results (1) RASSF1A mRNA was expressed in all normal tissues, but there was a high rate of loss of expression (53.2%) in lung cancer tissues. (2) The rate of loss of RASSF1A mRNA (70 4%) was significantly higher in patients with positive lymph node metastasis than those with negative lymph node metastasis (30 0%) (P 0 01). (3) The loss of RASSF1A mRNA was correlated with the stage of lung cancer (P <0.05). (4) The loss of RASSF1A mRNA expression was not associated with tumor histological type, tumor differentiation and smoking index (P> 0.05). Conclusion The lack of transcriptional expression of RASSF1A may be involved in the regulation of the carcinogenesis and development of lung cancer, which may affect its biological characteristics and prognosis.
