目的探讨唐山地区汉族人群急性脑梗死患者白细胞介素-6(IL-6)基因启动子-572C/G、-174C/G在血清中表达与脑梗死病情的关系。方法采用PCR-RFLP法检测IL-6-572C/G、-174C/G,酶联免疫法检测IL-6表达水平。结果脑梗死患者中IL-6水平明显高于对照组,IL-6、-572/G、174G/C基因型频率与等位基因频率与对照组比较也有显著差异。结论基因启动子-572G/C与脑梗死发病具有相关性,且可能为脑梗死发病的遗传易感基因,G等位基因个体通过促进IL-6表达而提高脑梗死患病风险。
Objective To investigate the relationship between the expressions of interleukin-6 (IL-6) gene promoter-572C / G and -174C / G in serum and the severity of cerebral infarction in Chinese Han population with acute cerebral infarction in Tangshan. Methods IL-6-572C / G and -174C / G were detected by PCR-RFLP and the expression of IL-6 was detected by enzyme-linked immunosorbent assay. Results The levels of IL-6 in patients with cerebral infarction were significantly higher than those in the control group. The frequencies of IL-6, -572 / G and 174G / C genotypes and allele frequencies were also significantly different from those in the control group. Conclusions The -572G / C gene promoter is correlated with the onset of cerebral infarction and may be a genetic predisposition to cerebral infarction. Individuals with G allele may increase the risk of cerebral infarction by promoting IL-6 expression.
