目的:用高效液相色谱法对经辐照灭菌的黄芩药材、黄芩药材粉末及未经灭菌黄芩药材中黄芩苷的含量进行测定。比较灭菌前后黄芩药材及其粉末中黄芩苷含量变化情况。方法:采用Accurasil-C18色谱柱(4.6 mm×250 mm,5μm);流动相甲醇-水-磷酸(47∶53∶0.2),检测波长280 nm,流速1.0 m L·min-1,柱温30℃,进样量10μL。结果:黄芩苷在0.163 2~0.816 0μg线性关系良好(r2=0.999 8),平均回收率为99.44%,RSD 1.6%。灭菌前后饮片与粉末中黄芩苷含量均有显著性变化。结论:以黄芩苷含量为指标考察,表明黄芩药材不适宜应用辐照灭菌法进行灭菌。
OBJECTIVE: To determine the content of baicalin in irradiated sterilized Radix Scutellariae Radix, Radix Scutellariae medicinal powder and non-sterilized Radix Scutellariae by high performance liquid chromatography. The changes of baicalin in Radix Scutellariae before and after sterilization were compared. Methods: Accurasil-C18 column (4.6 mm × 250 mm, 5 μm) was used. The mobile phase was methanol-water-phosphoric acid (47:53:0.2). The detection wavelength was 280 nm and the flow rate was 1.0 m L · min- ℃, injection volume 10μL. Results: The linearity of baicalin at 0.163 2-0.816 0 μg was good (r2 = 0.999 8). The average recoveries were 99.44% and RSD 1.6%. The content of baicalin in the tablets and powder before and after sterilization both had significant changes. Conclusion: The content of baicalin as an index, indicating that Scutellariae Radix is not suitable for sterilization by irradiation sterilization.
