Objective: To observe the protective effect of total flavonoids from Hemerocallis fulva on acute alcoholic liver injury in mice and to explore its possible mechanism. Methods: Sixty mice were randomly divided into normal group, model group, bifendate group and Hemerocallis fulva total flavonoids in low, medium and high dose groups. Acute alcoholic liver injury model was established by one-time intragastric administration of ethanol. To observe the effects of total flavones from Hemerocallis fulva on the activities of serum ALT and AST and the activities of superoxide dismutase (SOD), malondialdehyde (MDA), malondialdehyde (MDA) (GPX) and glutathione (GSH), and observed the pathological changes of liver tissue. Western blot was used to detect the expression of Bcl-2 (Bcl-2) -2), Bcl-2-related protein X (Bax) protein expression. Results: Compared with the model group, the total flavonoids of Hemerocallis fulva could significantly reduce the activity of ALT and AST (P <0.05), decrease the content of MDA in liver tissue and increase the activities of SOD, GPX and GSH (P <0.05, P <0.01). Pathological examination shows that, compared with the model group, hemerocallis total flavonoids, high dose group can significantly reduce the degree of hepatocyte degeneration and necrosis, improve the pathological changes of liver tissue. Western blot results showed that Bcl-2 protein expression increased (P <0.05, P <0.01); Bax protein expression decreased, Bax / Bcl-2 decreased (P <0.05, P <0.01). Conclusion: Flavonoids from Hemerocallis fulva have significant protective effect on alcohol-induced acute alcoholic liver injury in mice. The mechanism may be related to its antioxidation and regulation of the expression of Bcl-2 and Bax proteins.
