目的 研究胚胎小鼠骺板Ⅰ型胶原的表达情况及雌激素对其表达的影响。方法 采用免疫组织化学方法 ,研究体外培养胚胎小鼠长骨骺板静止区、增殖区和肥大区Ⅰ型胶原蛋白表达情况 ,同时观察10 -7、10 -8、10 -9mol/L雌酚酮对其表达水平的影响。结果 正常对照组骺板各区均有Ⅰ型胶原蛋白免疫组织化学染色阳性细胞。当培养基中雌酚酮浓度为 10 -7mol/L和 10 -8mol/L时 ,各区Ⅰ型胶原蛋白免疫组织化学染色阳性细胞面积与对照组相比均增加。当浓度降至 10 -9mol/L时 ,则与对照组差异不显著。随浓度升高 ,静止区和肥大区阳性细胞面积呈逐渐增加趋势。除 10 -7mol/L浓度下 ,静止区阳性细胞数与对照组相比增加外 ,各实验浓度下 ,各区阳性细胞数与对照组相比差异不显著。结论 在体外培养条件下 ,胚胎小鼠骺板静止区、增殖区和肥大区可表达Ⅰ型胶原蛋白 ,且表达水平可被雌激素上调
Objective To study the expression of type Ⅰ collagen in epiphyseal plate of embryonic mouse and the effect of estrogen on its expression. Methods Immunohistochemical method was used to study the expression of collagen Ⅰ in the quiescent zone, proliferative zone and hypertrophy zone of embryonic mice in vitro. At the same time, the effects of 10 -7,10 -8,10 -9 mol / L estrone on The effect of its expression level. Results There were type Ⅰ collagen immunohistochemical positive cells in the epiphyseal plate in normal control group. When the concentration of estrone in culture medium was 10 -7 mol / L and 10 -8 mol / L, the area of type Ⅰ collagen immunohistochemical staining positive cells in each area increased compared with the control group. When the concentration dropped to 10 -9 mol / L, the difference was not significant with the control group. With the increase of concentration, the area of positive cells in quiescent area and hypertrophic area increased gradually. Except for the concentration of 10 -7 mol / L, the number of positive cells in quiescent zone increased compared with the control group, and the number of positive cells in each zone was not significantly different from that of the control group at all experimental concentrations. Conclusion In vitro culture conditions, type I collagen can be expressed in the static zone, proliferative zone and hypertrophic zone in the epiphyseal plate of embryonic mice, and the expression level of which can be up-regulated by estrogen
